TransIT® In Vivo Gene Delivery SystemThe first system specifically designed for in vivo applications Features and Advantages
Product DescriptionThe TransIT® In Vivo Gene Delivery System is the first product specifically designed to introduce nucleic acids into live mice, allowing for organ-specific genetic studies that were previously impossible to perform on a routine basis. This product contains a unique polymer formulation that facilitates efficient delivery of exogenous DNA complexes into tissues of live mice without the use of potentially harmful viruses. The easy-to-use system consists of two components: a proprietary polymer that forms complexes with the DNA of interest, and a dilution/injection reagent for delivering the DNA into the animal. The DNA is injected into the tail vein of the mouse and target organs are assayed using simple reporter gene assays. The product yields a broad tissue-expression range. |

TransIT® -TKO: small interfering RNA (siRNA) Transfection ReagentFeatures and Advantages
Product Description
The novel TransIT® -TKO reagent enables highly efficient siRNA transfection with significantly reduced levels of cell damage as compared to cationic-liposome based transfection reagents. Transfections are most effective when carried out in complete growth media without a media change or serum addition. When complexed with even low levels of siRNA, TransIT® -TKO reagent successfully knocks out target gene expression in a variety of cell lines. Transgene expression can be silenced either when the siRNA is transfected before, concurrently or after the transfection of the expression vector. These unique features make TransIT® -TKO transfection reagent ideal for all siRNA-mediated gene-silencing studies. It is the only transfection reagent designed to efficiently deliver small interfering RNA to cells in vitro. TransIT® -TKO reagent, combined with the correct siRNA sequence against the target mRNA, can silence endogenous, stable or transient gene expression. |

| Knockdown of Luciferase Expression with TransIT-TKO® Reagent in Cos-7 Cells: Reporter plasmids expressing firefly and sea pansy luciferases were transfected into COS-7 cells using TransIT®-LT1 transfection reagent. After four hours various concentrations of anti-firefly luciferase siRNA (0, 0.05, 0.25, 0.75, 1.0, 5.0, 10.0 and 25 nM) were complexed with TransIT-TKO® Transfection Reagent and transfected into the COS-7 cells in their complete media. Twenty-four hours post-transfection, cell lysates were assayed for both firely and sea pansy luciferase expression using Promega's Dual-Luciferase Reporter Assay System. Points indicate percent firefly luciferase expression as compared to the untargeted sea pansy luciferase. |

The TransIT-TKO® Reagent is supplied as a dry film on clear plastic, sterile, 96-well plates which have flat-bottom wells suitable for cell culture. Each plate is supplied individually vacuum sealed in a foil pouch to maintain sterility and ensure effectiveness. Researchers can test up to eight different siRNA sequences in triplicate format, at four different levels of reagent, from L1 (lowest) to L4 (highest), using the TransIT-TKO® HTS-96 Titration Plate (MIR 2530), on their particular cell type. Once the optimal level has been determined based on the highest efficiency of transfection with the lowest cellular toxicity, additional plates can be purchased at the desired level. Transfections can be performed using a single TransIT-TKO® HTS-96 Plate or using a two step transfer method, where the TransIT-TKO® HTS-96 Plate is used as a reagent plate to form complexes and transfer to the plate of choice.

The TransIT®-Jurkat Transfection Reagent is specifically optimized to provide superior transfection efficiency in Jurkat cells. Jurkat cells (human T-lymphocyte; acute T-cell leukemia) have been inherently difficult to transfect, yet have remained a prevalent cell line in the immunological research field. The specificity of the TransIT®-Jurkat Transfection Reagent makes this product a desirable alternative to broad spectrum transfection reagents. The reagent provides many attributes of the trusted TransIT® Reagent line: high efficiency, simplicity of use, and reproducibility. Transfections with the TransIT® Reagents do not require media changes and can be carried out in serum-containing media. These significant features establish the TransIT®-Jurkat Transfection Reagent as the product of choice for transfecting Jurkat cells.
