Destaining BagsAMRESCO destaining bags efficiently remove Ethidium Bromide, Coomassie Blue, and other biological dyes from solution. The special adsorbent mixture retains dye molecules in a convenient bag for safe removal and incineration. Each bag will extract up to 5 mg of Ethidium Bromide from solution during an overnight treatment. |

How to use AMRESCO’s Destaining BagsThe Destaining Bags from AMRESCO are an acid/base mixed bed of resin, which are expected to absorb 1-5 mg of ethidium bromide from gel solutions and running buffers after gentle overnight agitation. The intent of these bags is to remove hazardous dyes like ethidium bromide from gel solutions for easy disposal. For example, if you have a 200 ml solution with 2 g/L of ethidium bromide, the solution contains 200 mg of dye. To remove all the dye from the solution would require (200/5 =) 40 Destaining Bags. If the intent is to remove Coomassie from a solution, there are several issues that need to be understood first. The Coomassie molecule is over 2 times as large as the ethidium bromide molecule, which means dye removal will not be as effective as with ethidium bromide removal. Also, Coomassie R-250 is dissolved in an acetic acid solvent. When the Destaining Bags are placed into this solution the resin becomes saturated with the acetic acid, resulting in slower and not as effective removal of the dye. Concentration removal is claimed only for ethidium bromide removal from solution. Use for removal of other dyes may require less or more bags and time for complete treatment.

SYBR® Green I
SYBR® Green I Nucleic Acid Stain is one of the most sensitive stains available for detecting DNA in agarose and polyacrylamide gels. Less than 20 pg of double-stranded DNA can be detected in a single band using 254 nm epi-illumination, black and white Polaroid® 667 print film, and SYBR® Green gel stain photographic filter. This sensitivity is 25 times greater than staining achieved using ethidium bromide under standard 312 nm transillumination. SYBR® Green I stain does not inhibit the activity of many common restriction endonucleases1,2 making it compatible with in-gel digestions and ligation techniques. Thus, the removal of SYBR® Green I stain is not necessary when sub-cloning stained DNA fragments isolated from gels. |

SYBR® Green II
SYBR® Green II RNA gel stain is an exceptionally sensitive dye for detecting RNA in electrophoretic gels. As little as 100 pg RNA or single-stranded DNA per band can be detected in a SYBR® Green II dye-stained agarose or polyacrylamide gel using 254 nm epiillumination, black and white Polaroid 667 print film and a SYBR® Green gel stain photographic filter.3,4 Even with 300 nm transillumination, as little as 500 pg RNA per band can be detected. In agarose/formaldehyde gels with SYBR® Green ll RNA gel stain does not interfere with transfer of RNA to filters or subsequent hybridization in Northern blot analysis as long as 0.1% - 0.3% SDS is included in prehybridization and hybridization buffers. SYBRC Green II stain is also compatible with amplification by PCR after SSCP analysis, SYBR® Green II stained bands can be excised out of the gel and used in cycle-sequencing.

Loading Dyes
Loading dyes are designed to optimize loading DNA, RNA, and proteins in acrylamide and agarose gels. Loading dyes serve three functions in electrophoresis. First, dyes impart color to the sample to facilitate the loading process. Second, loading dyes increase the density of the sample to ensure efficient sample distribution into each well. Third, the dyes themselves migrate independently from the samples, allowing the user to estimate the migration of nucleic acids or proteins.
