The Amplex ® Red reagent (10-acetyl-3,7-dihydroxyphenoxazine), a highly sensitive and stable probe for H2O2, is our best fluorogenic substrate for peroxidase. In the presence of horseradish peroxidase (HRP), the Amplex Red reagent reacts in a 1:1 stoichiometry with H2O2 to produce highly fluorescent resorufin. As H2O2 is produced in many different enzymatic reactions, the Amplex Red reagent allows researchers to detect the activity of many different enzymes.

AnaSpec offers nucleic acid stains, e.g. the minor groove binders such as DAPI and the Hoechst dyes.
DAPI emits blue fluorescence upon binding to AT regions of DNA (Exc./Em. max.: 358/461 nm). Although the dye is cell impermeant, higher concentrations will enter a living cell. DAPI is suitable for nuclei and chromosome counterstaining.
The blue fluorescent Hoechst 33342 dye (Exc./Em.max.: 350/461 nm) has been used widely for staining the nuclei of living cells. Hoechst dyes preferentially bind to AT regions, making them quite selective (but not specific) for DNA. The permeant Hoechst 33258 (Exc./Em.max.: 352/461 nm) has been successfully used for cell cycle studies.
The intercalating dye propidium iodide (PI) is a popular red-fluorescent nuclear and chromosome counterstain. Since propidium iodide is not permeant to living cells, it is commonly used to detect dead cells in a population. PI can be excited with mercury- or xenon-arc lamps or with the argon-ion laser, making it suitable for fluorescence microscopy, confocal laser-scanning microscopy, flow cytometry and fluorometry.
Actinomycin D is a non-fluorescent intercalator that exhibits high GC selectivity. Binding of actinomycin D to nucleic acids changes the absorbance of the dye.
Dihydroethidium is a chemically reduced ethidium derivative that is permeant to live cells. Dihydroethidium exhibits blue fluorescence in the cytoplasm. Many viable cells oxidize the probe to ethidium, which then fluoresces red upon DNA intercalation.

Aequorin is a calcium-sensitive photoprotein originally obtained from the jellyfish Aequorea aequorea. Since it has a high sensitivity to calcium ions, aequorin is widely used as a probe to monitor intracellular levels of free calcium. The aequorin molecule also contains coelenterazine as its chromophoric ligand which emits blue light when bound to calcium ions. We offer 5 coelenterazine products (cp, f, h, hcp and n) wich differ from each other in terms of luminescence wavelength, capacity, intensity, and half-rise time. They are widely used in applications such as calcium imaging, diagnostic immunoassays and high-throughput drug screening.

The cationic rhodamine based indicator Rhod-2, with fluorescence excitation and emission maxima at 552 nm and 581 nm, has been successfully used as selective indicator of mitochondrial calcium.
Indo-1 is an UV-light excitable ratiometric calcium indicator and is the preferred dye for flow cytometry, where it is more practical to use a single laser for excitation - usually the 351- 364 nm spectral lines of the argon-ion laser and monitor two emissions. The emission maximum of indo-1 shifts from ~475 nm in Ca2+-free medium to ~400 nm when the dye is saturated with Ca2+.

H2DCFDA is a cell-permeant indicator for reactive oxygen species that is nonfluorescent until the acetate groups are removed by intracellular esterases and oxidation occurs within the cell.
DiI and DiO are widely used as anterograde and retrograde neuronal tracers in living and fixed tissues and cells. DiI labeling does not appreciably affect cell viability, development or basic physiological properties. DiI-labeled cells have remained viable for up to four weeks in culture and up to one year in vivo.
