SYBR SafeTM DNA Gel Stain

was developed specifically for reduced mutagenicity - to be safer than ethidium bromide for staining DNA in agarose or acrylamide gels. SYBR Safe stain is not only less mutagenic than ethidium bromide, but SYBR Safe stain's detection sensitivity is better than that of ethidium bromide. SYBR Safe stain comes as a premixed solution that can be used just like an ethidium bromide solution, either in the gel during the electrophoresis run or as a post-run stain. Bound to nucleic acids, the green-fluorescent SYBR Safe stain has fluorescence excitation maxima at ~280 and ~502 nm, and an emission maximum at ~530 nm. SYBR Safe DNA gel stain is also available as 4 L (S-33101) of ready-to-use solution. The SYBR Safe DNA Gel Stain Starter Kit (S-33110) includes 1 l of SYBR Safe DNA gel stain and one photographic filter (S-37100).

Electrophoretic Mobility-Shift Assay (EMSA) Kit

Fast, easy and quantitative, our new Electrophoretic Mobility-Shift Assay (EMSA) Kit (E-33075) enables you to detect nucleic acid and protein in the same gel without using a radioactive label. This kit uses two fl uorescent dyes for detection — SYBR Green EMSA stain for RNA or DNA and SYPRO Ruby EMSA stain for proteins. Because the gels are stained postelectrophoresis, there is no need to label the DNA with a radioisotope, biotin or a fluorescent dye before the binding reaction, and therefore there is no possibility that the label will interfere with protein binding. Staining takes only 20 minutes for the nucleic acid stain and a few hours for the protein stain, yielding results much faster than radioactive labeling (which may require multiple exposure times) or chemiluminescence-based detection (which requires blotting and multiple incubation steps). This kit also makes it possible to perform ratiometric measurements of nucleic acid and protein in the same band, providing more detailed information on the binding interaction. Both stains can be detected using a standard UV transilluminator or a laserbased scanner. Digital images can easily be overlaid for a two-color representation of nucleic acid and protein in the gel.

Titration of lac operator DNA with lac repressor protein.

Titration of lac operator DNA with lac repressor protein. Increasing amounts of lac repressor protein were mixed with 40 ng of lac operator DNA, incubated for 20 minutes and then separated on a 6% nondenaturing polyacrylamide gel. The gel was stained with SYBR Green EMSA stain (green) followed by SYPRO Ruby EMSA stain (red). After each staining, the image was documented using an FLA-3000 laser-based scanner (Fuji) and the digital images were pseudocolored and overlaid. Yellow indicates DNA-protein complexes that were stained by both stains.

SYBR Gold nucleic acid gel stain is the most sensitive gel stain available for detecting dsDNA, ssDNA and RNA in native, glyoxal, formaldehyde or urea gels. Use with UV transilluminators, the Dark Reader, or laser scanners. 500 µL stains 50 minigels.

SYBR® Green I

SYBR® Green I nucleic acid gel stain is an exceptionally sensitive nucleic acid gel stain that has bright fluorescence when bound to dsDNA and low background in gels, making it ideal for detecting dsDNA in gels using laser scanners or standard UV transilluminators. SYBR® Green I nucleic acid stain has also found uses in capillary electrophoresis, real-time PCR assays and bandshift assays.

Quant-iTTM Assay Kits

The Quant-iTTM Broad-Range DNA Assay Kit makes DNA quantitation easy and accurate. The kit provides concentrated assay reagent, dilution buffer, and pre-diluted DNA standards. The assay is highly selective for double-stranded DNA over RNA, and in the range of 2-1000 ng of DNA, the fluorescence signal is linear. A Quant-iTTM DNA Assay Kit with a range of 0.2-100 ng of dsDNA is available (Q33120).

The Quant-iTTM RNA Assay Kit makes RNA quantitation easy and accurate. The kit provides concentrated assay reagent, dilution buffer, and pre-diluted RNA standards. The assay is highly selective for RNA over double-stranded DNA, and in the range of 5 - 100 ng of RNA, the fluorescence signal is linear.

Our Quant-iTTM OliGreen ssDNA Assay Kit contains an ultrasensitive, green-fluorescent nucleic acid stain for quantitating oligonucleotides and single-stranded DNA (ssDNA) in solution. In addition to the dye, the kit also includes buffer and an oligonucleotide standard. The OliGreen reagent is also available as a stand alone product (O7582).

The Quant-iTTM PicoGreen dsDNA Assay Kit contains our Quant-iTTM PicoGreen dsDNA reagent as well as buffer and DNA standards. The Quant-iT PicoGreen reagent is highly selective for double-stranded DNA over RNA, and in the range of 0.2 - 200 ng of DNA, the fluorescence signal is linear. This reagent is also available separately (P7581).

The Quant-iTTM RiboGreen RNA Assay Kit contains Quant-iT RiboGreen RNA reagent as well as buffers and RNA standards. Quant-iT RiboGreen RNA reagent is one of the most sensitive detection dyes for the quantitation of RNA in solution, with linear fluorescence detection in the range of 1-200 ng of RNA.
