LDH Cytotoxicity Detection Kit

Features and Advantages
  • Safe: No radioactive isotopes are used.
  • Accuracy: Assay results obtained with this kit strongly correlate to the number of damaged cell.
  • High sensitivity: Low cell numbers, such as 0.2 - 2 x 102 cells/ well, can be available for detection.
  • Fast: A large number of samples can be processed simultaneously by using a multiwell-ELISA reader. It takes only 0.5 - 1 hour for measurement.
  • Simple: No need for prelabeling and washing steps. As the kit does not employ radioactive isotopes, no disposal and radiation safety operations or paperwork are required.

Description:

Lactate dehydrogenase (LDH) is a stable cytoplasmic enzyme which is present in most cells. It is released into the cell culture supernatant upon damage of the cytoplasmic membrane. The culture supernatant is incubated with the reaction mixture supplied in the kit. Then, the LDH activity is measured in an enzymatic test. (See Figure).

Principle: In the first step NAD+ is reduced to NADH/H+ by the LDH-catalyzed conversion of lactate to pyruvate. In the second step, the catalyst (diaphorase) transfers H/H+ from NADH/H+ to the tetrazolium salt which is reduced to formazan. An increase in the number of dead or plasma membrane-damaged cells leads to an increase of the LDH enzyme activity in the culture supernatant. This leads to color change from pale yellow to red. Formazan exhibits an absorption maximum at 500 nm, whereas the tetrazolium salt INT shows no significant absorption at this wavelenth.

Premix WST-1 Cell Proliferation Assay System


Features:

  • Ease of use: Supplied as a ready-to-use, sterile solution; short reaction time; the entire assay is performed in one microtiter plate; no need for washing, harvesting or formazan dye solubilization; processing a large number of samples is possible by using a multiwell-ELISA reader; no isotope disposal and radiation safety paperwork.
  • Flexibility: Plates can be read and returned several times to the incubator for further color development.

Description:
The PreMix WST-1 enables to measure the cell proliferation and cell viability with colorimetric assay, and bases on the cleavage of tetrazolium salts by mitochondrial dehydrogenase in viable cells.

Principle: Total activity of this mitochondrial dehydrogenase in a sample rises with the increase of viable cells. As the increase of enzyme activity leads to an increase of the production of formazan dye, the quantity of formazan dye is related directly with the number of metabolically active cells in the medium. The formazan dye formed by metabolically active cell can be quantitated by measuring its absorbance by ELISA reader. The absorbance of formazan dye solution is in direct proportion to the number of viable cells.

Description:

GPDH catalyzes the reversible reaction between dihydroxyacetone phosphate and glycerol 3-phosphate with NAD as coenzyme. The enzyme activity has been used as an index for monitoring the fat synthesis. This kit can be used for screening of differentiation repressors and for studies of differentiation mechanism on the experimental system performing differentiation of primary or established cell-lined progenitor fat cells into fat cells.

Principle:
Dihydroxyacetone phosphate + NADH + H+ arrow Glycerol 3-phosphate + NAD+
The decrease of NADH in absorbance at 340 nm is measured and the activity of GPDH is calculated.
