 Monoclonal antibodies against human cathepsins and cystatin A

Cathepsin L is a member of the papain family of closely related cysteine proteinases and it seems to be involved in a variety of disorders in which the pathological destruction of the extracellular matrix is one of the main characteristics. The numbering of the amino acids of the recognized epitopes is according to that of the human procathepsin L sequence. The sequences of the mature single and two chain enzymes are beginning at amino acid 97 of the procathepsin L sequence. The heavy chain (24 kDa) of mature two chain cathepsin L comprises amino acids 97 to 271, the light chain (4 kDa) those of amino acids 275 to 316.
Cathepsin W (lymphopain) is a recently described cysteine proteinase which is predominantly expressed by CD8+ -T-lymphocytes and natural killer cells. Due to sequence similarities between the propeptides of cathepsin W and cathepsin F, both enzymes may form a new cathepsin subgroup. Cystatin A (acid cysteine proteinase inhibitor, ACPI; Stefin A) is a natural inhibitor of cysteine proteinases. It seems to be important in the regulation of the intra- and the extracellular proteolytic potential of many cells and was recently shown to be involved in controlling apoptosis.
