Cytokines play an important role in the regulation of immune functions (i.e. immune responses) mediating the communication between various hematopoietic cells and influencing lymphocyte development.
Recently, new cytokines have been discovered designated IL-21 and IL-22. The latter is an IL-10 homologue which is produced by activated T-cells. It may be involved in inflammation because it induces the production of acute-phase reactants in vitro and in vivo.
IL-22 binds to and signals through CRF2-4, a member of the class II cytokine receptor family which also includes IL-10R. IL-22R, a new member of the interferon receptor family, functions as a second component together with CRF2-4 to enable IL-22 signaling.
IL-21 is an exciting new member of the cytokine family which influences lymphoid differentiation and is probably used by T-cells to stimulate effector functions in an autocrine and paracrine fashion. It has structural homologies with IL-15, IL-4 as well as IL-2 and has been found to have potent effects on all classes of lymphocytes (B, T and natural killer cells). IL-21 may substantially increase the cytotoxic activity of mature natural killer cells.
The IL-21 receptor shows significant homologies to the IL-2, IL-4 and IL-9 type I receptors. It is expressed by activated T-cells. A further new member of the type I cytokine receptor family that plays a part in the T-cell differentiation is designated TCCR (T-cell cytokine receptor).
