Luciferase activity in liver cells 24h post-injection

TransIT®-EE Solution Leads to Enhanced Luciferase Expression in the Liver.
TransIT®-QR or TransIT®-EE Delivery Solutions were used to deliver 10 µg of a luciferase expression vector to mice using the hydrodynamic delivery tail vein injection procedure. Twenty-four hours post-injection the livers were harvested and assayed for luciferase activity.

TransIT®-EE Solution Leads to Enhanced Luciferase Expression in a Variety of Organs.
TransIT®-QR or TransIT®-EE Delivery Solutions were used to deliver 10 µg of a luciferase expression vector to mice using the hydrodynamic delivery tail vein injection procedure. Twenty-four hours post-injection the livers were harvested and assayed for luciferase activity.

NOTE: Following gene delivery via a tail vein injection, the highest level of transgene expression is found in the liver, with reduced levels of expression found in the spleen, lung, heart and kidneys.

Description

  • The TransIT®-EE (Enhanced Expression) Hydrodynamic Delivery Solution is designed specifically for the delivery of DNA expression vectors into laboratory mice using the hydrodynamic tail vein injection procedure. This formulation is designed to produce high-level gene expression from the delivered DNA in the liver with significant but reduced levels of expression in the spleen, lungs, heart, and kidneys. This formulation offers the additional benefit that the injected mice demonstrate enhanced expression post-injection compared to animals injected using normal saline.

    The expression level obtained using this solution is approximately 2-3 fold greater than the level of expression obtained after delivery of DNA using the TransIT®-QR Hydrodynamic Delivery Solution. The TransIT®-EE Delivery Solution is not optimal for the delivery of siRNA and does not promote the same quick recovery as the TransIT®-QR Delivery Solution.

    The Delivery Solution is certified RNase-, DNase-, and endotoxin-free and provided in a ready-to-use 1X format.
