| Description: | General Drug Target ׀ The primary biological function of bacterial DNA topoisomerase IV (the ParC-parE complex) enzymes is DNA decatenation. Bacterial Topo IV enzymes also have DNA relaxation and ATPase activities. Some inhibitors such as ciprofloxacin cause DNA damage by DNA cleavage (topoisomerase poisons). ProFoldin offers all the assays for various biological functions of DNA topoisomerase IV enzymes and the DNA cleavage assay of topoisomerase poison inhibitors. The DNA decatenation assays are carried out in a simple spin-column format or a high throughput 96-well plate format. This product includes 150 ul of 10 x assay buffer (Buffer T4), 105 ul of 10 x concatenated DNA, 30 ul of 50 x ATP (10 mM),120 ul of 0.4 M EDTA, 160 ul of 20 x fluorescence dye and 20 spin columns for 20 DNA decatenation assays. The assay buffer is optimized for bacterial topoisomerase IV. Bacterial topoisomerase IV enzymes are provided separately. |
