| Description: | Anticancer Drug Target ׀ Human topoisomerase I relaxes supercoiled plasmid DNA to form relaxed DNA. The Human DNA topoisomerase I assay is based on the principle that the supercoiled DNA and relaxed DNA yield different fluorescent intensity when interact with fluorescence dye H19. The relaxed DNA suppresses the fluorescent intensity much more than the supercoiled DNA. Therefore, when the supercoiled DNA is converted into its relaxed form, the fluorescent signal decreases. The change of fluorescence intensity is used to measure the relaxation reaction. This product includes 4800 ul of 10 x Buffer HT1, 4050 ul of 10 x supercoiled plasmid DNA, 40 ul of 1000 x human topoisomerase I enzyme (500U/ul), 170 ul of 1500 x Dye H19 and 30 ml of 10 x H19 dilution buffer. This product includes all the reagents for 1000 DNA relaxation assays in a 96-well plate format or 2000 assays in a 384-well assay format. |
